Hope Scooby, Shaggy and Daphne will be the main characters of it and they will be in character without the annoying personalities WB gave them.

Shaggy and Scooby are the main characters in pretty much everything even if it's to the detriment of the show i.e. when they are used as filler - just running about and yelling. How about giving all of the gang equal time and well written characters?

Yes equal treatment is needed and it should have all 7 members of Mystery Inc together for once, i kinda wonder what Flim thinks about Fred and Velma...

I suspect it will be 'the gang ' as has been in most of the shows i.e. Fred, Daphne, Velma, Shaggy and Scooby. It is possible, but IMO not probable that you will get others in . After all you have a long list of candidates to include; Scooby Dum, Scooby Dee, Madelyn Dinkley, Daphne's relatives, Shaggy's uncle etc.

^They'd probably go with Flim and Scrappy before the other relatives if they did add new members seeing as they lasted longest, Flim had a full season (13 episodes) and Scrappy had ten years, it'd also be less confusing than Dee and Dum. I mean three Scooby Dooby calls in one series/film oh my! Still though just the normal status quo is mist likely.

The decision would be based on what fits with the story, not longevity, also on what they perceive to fit well with the audience. If SCOOB is for a wider audience they will go with what is in the wider cultural 'pool'. Most will remember 'those meddling kids, someone loosing their glasses, someone being 'danger prone' and a lanky guy and his dog eating food, lots and lots of food. If you are appealing to people outside of the fandom then that is what you construct your story around, not a particular section of the fan-base. There are things I'd like to see in a file, that you want to see in a film, are probably diametrically opposed to each other, doesn't mean they would make movies that would sell well.

The best chance we have for Scrappy is him being featured in a DTV in the hopes it will change audience's impressions of him.

I doubt they are going to get too inclusive in only the first movie. That is going to take some time to bring in the franchises/characters that need a little more nudging. I sincerely doubt anything not main stream is going to be directly appearing in S.C.O.O.B. if anything they will probably work in references for future films.
Besides we still have no real conclusive numbers of exactly how well some of this stuff does. By comparison we know it's clearly doing better than all the other WB animated brands, but by how much? Enough they can afford to market it as nerdy as the dtv's are? Good but needs a wider audience to sell a theatrical release? Things like this make a world of difference and even those of us who are kind of obsessed on it, don't really have a solid answer yet.

I think we're still trying to get Daphne right in the dtvs. Everyone else is fine, but Daphne they keep changing her personality so much it's hard to keep track, just do like Fred and declare her WNSD persona the defacto one (it's been her best one really.) Now they have tried Flintstones and Jetsons for HB with illusions for Johnny Quest in Scooby. Hopefully Johnny can get an adventure film soon. Also with this SCOOB film i would say since WB started with Supes and Bats in a partial N52 setting for DC, i would base the new film partially on The Scooby Team ups like a little nod here or there to the specific HB cartoons (aired or not as we know that the junior JLA aka the teen titans do have HB designs/counterparts) starting with maybe Quest or Super Friends or the Blue Falcon, probably Quest since Quest labs is mentioned about every so often. The DC movies and shows do this by saying "oh that nut in Gotham?" and the like. If you are building a universe you need to state who's all in that universe so WB could just say something to the effects of "You mean those four who once had twin sidekicks?" for a super friends nod.

I sincerely doubt they will cross pollinate any regular DC things into this film. Especially after there DC live action universe project has gotten as much flack as it has already. Which is one that took a gamble on utilizing two stories that even DC fans are mixed about. I think they will want to separate them apart for this. As after all back catalog DC has taken a nose dive in releases now too.
They have a bunch of super heroes of their own to deal with if they want to go that route. Which I bet is something Future Quest is being watched to see how much of a reaction it can stir up.

i doubt we'll see scooby and the gang run into batman or anything like that. might see him meet johnny quest though.

Considering that Hanna-Barbera has it's own earth in the DC Multiverse (as DC does all the books) called earth 1A then we might. Remember that DC's mainstream JLA is not the only JLA that exists and DC loves to do multiverse stories. Also the general public would want to see either in the hbu or a dtv a new Scooby-Bat/Scooby-JLA teamup. DC has a pattern Main bat, main jla, dtv episodes one out of continuity film with the scooby stuff always following the bat and out of continuity film.
