I've heard of cases where the older twin is referred to as the big sibling and the younger as the little one. Madelyn and Velma seem to be very close in age.

No, they are sisters. That's what they say in Abracadabra Doo. What's your vote russm? It's a no. At no point in the film is there any indication that they are twins. Velma refers to Madelyn as her little sister while Madelyn calls Velma her big sister, something I contend that twins would not do.

twins is very unlikely. i sense theres is at least a years age difference between velma and Madelyn. twins would be born within hours, minutes or seconds of each other. this is not the case with these two.

Good point. I am a twin (The older one) And my sister HATES being referred as the little one. Although people do call us by older and younger. (It helps that she really does look younger then me) I suppose it's possible Madelyn wouldn't mind, maybe, maybe not.
